Alteration of speed-of-sound by fixatives and tissue processing methods in scanning acoustic microscopy

The elasticity of biological tissues is one the physical characteristics and has attracted attention as a clinical diagnostic parameter. can be determined on microscopic scale with speed sound (SoS) measurements using acoustic microscopy. In SoS measurements, thin-sliced section attached to glass slide in same manner light specimen. There are two main methods for preparing thin sections: paraffin-embedding frozen-section. frozen-section method requires fewer processing steps from sectioning measurement considered reduce artifacts sample compared method. Both need fixatives keep tissue structures. Many reports frozen sections focused soft relatively high protein contents. this study, we four types organs (brain, heart, liver, kidney) prepared different (paraffin-embedding frozen-section) chemical (formalin, Karnovsky fixative (KF) 0.5% 2.0% glutaraldehyde, ethanol). heart liver samples KF showed good agreement reported values raw samples. For fixed KF, increased glutaraldehyde concentration 2.0%. A brain tumor was processed 0.5%, significantly higher than that non-tumor area. results confirmed it possible measure low contents appropriate fixatives.
